No, the Osmo Carrying Cases are not required in order to use any of the Osmo products.
However, the Osmo Carrying Cases are the easiest way to keep your Osmo organized and protected on the go! Both the Osmo Large Carrying Case and the Osmo Small Carrying Case are specifically designed to fit the new Osmo Base for iPad plus several Osmo games (depending on stackable storage container size).
